Mechanism of Action

Humanin is a 21-amino acid peptide encoded within the 16S rRNA gene of mitochondrial DNA, representing a class of mitochondrial-derived peptides (MDPs). It binds to the humanin receptor complex (CNTFR/WSX-1/gp130 or formyl peptide receptor) and activates JAK2/STAT3 signalling. This activation suppresses neuronal apoptosis induced by Alzheimer-associated insults (amyloid-beta, presenilin mutations), promotes insulin sensitivity via IGF-1 signalling, and reduces oxidative stress. Circulating humanin levels are higher in long-lived individuals and centenarian offspring.
