Mechanism of Action

MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA Type-c) is encoded in the mitochondrial 12S rRNA sequence. It translocates from mitochondria to the cytoplasm and nucleus in response to metabolic stress. In the cytoplasm, it inhibits the folate cycle (specifically the MTHFD2 enzyme), reducing purine synthesis and activating AMPK via AICAR accumulation. Nuclear MOTS-c binds to ARE (antioxidant response elements) and regulates stress response and metabolic genes. Plasma MOTS-c levels decline with age and are elevated by exercise.
