Mechanism of Action

PE-22-28 is an optimised 7-amino acid analogue of spadin, a naturally occurring TREK-1 channel blocker. TREK-1 is a two-pore-domain K+ channel (K2P) that hyperpolarises neurons when open. By blocking TREK-1, PE-22-28 increases neuronal excitability in hippocampal and cortical circuits and promotes BDNF-dependent neurogenesis in the dentate gyrus - a mechanism distinct from classic monoamine antidepressants.
