FOXO4-DRI is a retro-inverso peptide analog of a FOXO4 alpha-helix that interferes with the interaction between FOXO4 and p53. In senescent cells, FOXO4 binds p53 in the nucleus and prevents p53 from entering the cytoplasm to trigger apoptosis, keeping senescent cells alive and metabolically active (secreting the SASP inflammatory cytokine profile). FOXO4-DRI competitively disrupts this interaction, releasing p53 to translocate to the mitochondria and initiate apoptosis specifically in FOXO4-overexpressing senescent cells.
