Mechanism of Action

NNMT consumes S-adenosylmethionine (SAM) to methylate nicotinamide, producing 1-methylnicotinamide and reducing cellular SAM and NAD+ availability. 5-Amino-1MQ is a small molecule inhibitor of NNMT, blocking this consumption pathway. By inhibiting NNMT, the compound raises NAD+ and SAM levels simultaneously, activating sirtuins (particularly SIRT1) and supporting methylation reactions. In fat cell models, NNMT inhibition reduces adipocyte differentiation and lipid accumulation.
