Mechanism of Action

Botulinum toxin type A (BoNT-A) is a 150 kDa zinc metalloprotease produced by Clostridium botulinum. It binds presynaptic nerve terminals via the heavy chain (HC), undergoes endocytosis, and the light chain (LC) is translocated into the cytoplasm where it cleaves SNAP-25 between Gln197 and Arg198. This irreversibly disrupts SNARE complex assembly, blocking ACh vesicle fusion and neuromuscular transmission until new SNAP-25 is synthesised over 3-4 months.
