Mechanism of Action

Melatonin (N-acetyl-5-methoxytryptamine) is synthesised from serotonin in pinealocytes during darkness, with synthesis controlled by the suprachiasmatic nucleus (SCN). It binds MT1 and MT2 receptors (GPCRs) in the SCN to suppress the alerting signal and shift circadian phase. It is also a direct free radical scavenger, protecting mitochondrial DNA and cristae from oxidative damage through a receptor-independent mechanism.
